| Summary, etc. | A simple, specific and sensitive high pressure liquid chromatographic method for the determination of rifampicin in human plasma of Filipino male and female healthy volunteers was developed and validated<br/>This method utilizes a direct precipitation separation technique and ultraviolet detection monitored at 337 nm. Dipyridamole is used as the internal standard. Rifampicin and the internal standard were separated using a Bakerbond narrow pore octyl (C8, 5um, 120 A, 4.6 x 250 nm) reversed-phase column with 10 mM phosphate buffer (pH 5.2), acetonitrile, and tetrahydrofuran (65:35:10) as the mobile phase. The retention time was 7.7 minutes for rifampicin and 14.5 minutes for dipyridamole. Results showed a linear response in the concentraion range of 0.5 to 20 mg/L (r=0.99925). The limit of quantitation is 0.5 mg/L. Both intraday and interday accuracy and precision data showed good reproducibility.<br/>The precision, accuracy, linearity, specificity, and sensitivity of the method were well-within the range of clinical utility and meet the requirements for bioavailability, bioequivalence, and pharmacokinetic studies.(Author) |
